Transport and Metering of Beads
Sepharose 4B beads were dyed green and blue to enhance visibility.
3.1.8.
1. Blue beads are metered by flow
reversal that packs them towards
the yellow plug and into the
groove of the rotor.
2. Forward flow transfers beads into
flow cell, where they are retained
by plug #2.
3. Beads are removed by
flow reversal, the valve is
turned and the beads are
flushed to waste.
The sequence is repeated
with green beads. Note
the absence of carryover.
Indeed, no bead is left behind!
